Changes in Mitochondrial Protein Patterns during Adventitious Root Formation of Gladiolus ¢¥ Topaz ¢¥ Callus


Abstract
Calli induced from cormel tissue of Gladiolus ¢¥Topaz¢¥ were subcultured on MS medium containing 0.5 §·¡¤L^(-1) 2,4-D. The callus was then transplanted to 2,4-D-free MS medium for adventitious root formation. Adventitious root formation was observed 7 days after transfer to the MS medium. Isocitrate dehydrogenase activities during adventitious root formation were the highest at root primordia-forming callus cultured for 6 days. The protein patterns of mitochondria during adventitious root formation were observed by isoelectric focusing-sodium dodecyl sulfate polyacrylamide gel electrophoresis (IEF-SDS¡¤PAGE). Approximately 130 polypeptides were separated by IEF-SDS¡¤PAGE and quantitative changes were observed in 6 polypeptides having molecular weights of 33 to 67kDa. Especially, several polypeptides increased in amount at root primordia forming callus and newly synthesized polypeptides were not observed.
